Quantitative lipidomics and standardization

Quantification is an essential task in comprehensive lipidomics studies challenged by the high number of lipids, their chemical diversity and the great dynamic range (up to 7 orders of magnitude) of the lipidome. Based on the labeling of yeast, we developed Lipidome Isotope Labeling of Yeast (LILY).

LILY is a resource-saving alternative to organic synthesis, as hundreds of lipids can be produced in parallel. It has been successfully used to improve relative and absolute quantification in yeast and human plasma. Our current research focuses on the applicability of the LILY approach to further sample matrices and increasing the number of lipid species by producing LILY standards optimized for different lipid classes.

Current topics:

  • Fractionation of 13C lipid extracts by super-critical fluid chromatography
  • Optimization and applications of LILY
  • Quantitative lipidomics
